Suppose I sign up with IB, and I'm doing either an intra-day strategy or one that can hold position a few days. For the intraday strategy, would it suffice to turn on WL8 before 930am EST and shut it off after 4pm EST? But if I do a strategy that holds a position overnight or for a few days, would I have to leave WL(and my computer) turned on the whole time to ensure it's sending the signals? TY.

If you're trading regular hours and holding many days, you don't have to leave anything running overnight. You only need to make sure that your strategies, when reinitialized the next day, are hypothetically holding the positions so that the exits will be managed. This may depend on how much data you use to initialize the strategy.

IB's TWS (or Gateway) must be restarted once each day. You can make that an automatic restart in the TWS Settings. You shouldn't assume WL8 will connect again after the TWS restart, so you should make sure that it does. Generally, it's a good idea to restart WL8 after the TWS restart. It will connect again automatically. You can even program the Task Scheduler to start WealthLab and open a specified Workspace at a specific time. (User Guide > Workspaces)
